Role of Perm1 in heart and skeletal muscle.
We have previously identified a novel mitochondrial regulator, Perm1 (PGC-1 and ERR induced regulator, muscle 1), in skeletal muscle cells (Cho et al., 2013). We have shown that Perm1 promotes mitochondrial biogenesis in the heart and skeletal muscle (Cho et al., 2016, 2019, 2021). Importantly, we found that Perm1 protects the heart from pressure overload by promoting oxidative metabolism (Tachibana et al., 2023). Currently, we are studying the mechanisms by which Perm1 modulates mitochondrial biogenesis and function and the role of Perm1 in the onset of heart failure and skeletal muscle dysfunction. We are also studying the role of Perm1 in exercise adaptation and whole-body metabolic homeostasis.

Obscurin, Obsculin linke 1, and metabolic regulation.
Obscurin (Obs) and Obscurin-like 1 (Obsl1) are giant sarcomeric proteins expressed in the heart and skeletal muscle and play a key role in muscle contractile function. In collaboration with Dr. Stephan Lange (UCSD, Aarhus University (Denmark)), we found that cardiac-specific Obs and Obsl1 double knock-out mice show mitochondrial abnormality and metabolic dysfunction in the heart. Currently, we are studying the mechanism by which Obs and Obsl1 modulate mitochondrial and metabolic functions.
